Tocotrienols and Vitamin E Health Topic

Vitamin E is composed of two structurally similar compounds: tocopherols (TPs) and tocotrienols (T3). A growing body of evidence suggests that in addition to its antioxidative and pro-apoptotic functions, tocotrienols possesses a number of anticancer properties that make it superior to tocopherols.2

  2. Tocotrienol As A Potential Anticancer Agent

Vitamin E is composed of two structurally similar compounds: tocopherols (TPs) and tocotrienols (T3). A growing body of evidence suggests that in addition to its antioxidative and pro-apoptotic functions, T3 possesses a number of anticancer properties that make it superior to TP. These include the inhibition of epithelial-to-mesenchymal transitions, the suppression of vascular endothelial growth factor tumor angiogenic pathway and the induction of antitumor immunity. More recently, T3 has been shown to have chemosensitization and anti-cancer stem cell effects.

  3. Tocotrienols: Inflammation and Cancer

Inflammation has long been associated with the development of cancer. The distinct characteristics of cancer cells to proliferate, metastasize, evade apoptotic signals, and develop chemoresistance have been linked to the inflammatory response. Due to the involvement of multiple genes and various pathways, current drugs that target single genes have not been effective. Natural products target multiple genes and therefore have better success compared to drugs. Tocotrienols, the potent isoforms of vitamin E, are such a natural product.

  4. Tocotrienol's Fight Cancer By Targeting Multiple Cell Signalling Pathways

Tumor cells acquire [multiple] properties due to the dysregulation of multiple genes and associated cell signaling pathways, most of which are linked to inflammation. Drugs that target a single gene product are unlikely to be of use in preventing or treating cancer. Moreover, targeted drugs can cause serious side effects. There is an urgent need for safe and effective [multitargeted]drugs. "Mother Nature" produces numerous such compounds that regulate multiple cell signaling pathways, are cost effective, exhibit low toxicity, and are readily available. Tocotrienol, a member of the vitamin E family, has exhibited anticancer properties.

  5. Tocotrienol-Rich Fraction of Palm Oil Induces Cell Cycle Arrest and Apoptosis Selectively in Human Prostate Cancer Cells

Tocotrienol-rich fraction (TRF) treatment resulted in significant decreases in cell viability and colony formation in all three prostate cancer cell lines. TRF treatment resulted in significant apoptosis in all the cell lines. These results suggest that...TRF is capable of selectively inhibiting cellular proliferation and accelerating apoptotic events in prostate cancer cells. TRF offers significant promise as a chemopreventive and/or therapeutic agent against prostate cancer.

  6. Tocotrienol: The Natural Vitamin E to Defend the Nervous System?

Vitamin E is essential for normal neurological function. Orally supplemented vitamin E reaches the cerebrospinal fluid and brain. The current findings point towards tocotrienol as a potent neuroprotective form of natural vitamin E.

  7. Clinical Trial: Tocotrienol Administered to Subjects With Resectable Pancreatic Exocrine Neoplasia

Vitamin E Delta-tocotrienol is being investigated for the prevention and treatment of pancreatic tumors. To characterize the safety and tolerability of Vitamin E delta-Tocotrienol when orally administered at up to 5.6 times the predicted biological effective dose (1600mg twice daily) for 14 consecutive days.

  8. Vitamin E delta-Tocotrienol Augments the Antitumor Activity of Gemcitabine and Suppresses Constitutive NF-kappaB Activation in Pancreatic Cancer

Tocotrienols are a promising class of anticancer compounds that inhibit the growth and survival of many cancer cells, including pancreatic cancer. Here, we show that tocotrienols inhibited NF-kappaB activity and the survival of human pancreatic cancer cells in vitro and in vivo. Delta-tocotrienol augmentation of gemcitabine activity in pancreatic cancer cells and tumors is associated with significant suppression of NF-kappaB activity and the expression of NF-kappaB transcriptional targets

  9. Clinical Trial: Vitamin E delta-Tocotrienol Single Dose in Healthy Subjects

The investigators preclinical studies indicate that delta-tocotrienol possesses the most potent antitumor activity against pancreatic cancer. It is believed that this micronutrient may have a role in the prevention of pancreatic cancer in healthy participants who are at increased risk of developing the disease.

  10. Vitamin E delta-Tocotrienol Levels in Tumor and Pancreatic Tissue of Mice after Oral Administration

In this report, we investigated the pharmacokinetics, tumor and pancreatic tissue levels, and toxicity of delta-tocotrienol in mice because of its anti-tumor activity against pancreatic cancer. This concentration was observed with the oral dose (100 mg/kg) of delta-tocotrienol which inhibited tumor growth by 80% in our previous studies. Interestingly, delta-tocotrienol was 10-fold more concentrated in the pancreas than in the tumor. We observed no toxicity due to delta-tocotrienol.

  11. Gamma-Tocotrienol Assists Bone Strength

Vitamin E, and in particular the form known as gamma tocotrienol, helps build stronger bones with better biomechanical strength. Studies showed that tocotrienols reduce inflammation in bone that leads to bone loss, can offset the harmful effects of steroid use on bones, and assist in the process of normal bone calcification.

  12. Tocotrienols Protect Against the Sun's UVB Radiation

Your skin has the ability to naturally activate its own defense system against UVB radiation. To do this it requires antioxidants within your skin to coordinate an optimal response. Gamma tocotrienol is able to regulate gene signals in skin to reduce damage caused by UVB radiation. Researchers showed that gamma tocotrienol reduced multiple inflammatory signals following UVB radiation exposure. When individuals take antioxidants like gamma tocotrienol on a regular basis then it accumulates in cell membranes and the skin, where it can help you enjoy the sun with less risk for adverse effects from UVB radiation.

  13. Gamma Tocotrienol Protects Against Radiation Damage

Excessive radiation exposure damages DNA, especially DNA relating to the system in our bone marrow that produces all the red and white blood cells that are vital for survival. In the current experiment mice were exposed to non-lethal amounts of whole body radiation. Stem cell colonies were 80% - 86% maintained in the gamma tocotrienol treated mice, while they were 50% reduced in controls. A detailed analysis of the bone marrow showed that gamma tocotrienol maintained the regenerative integrity of bone marrow cells.

  14. Health Benefits of Tocotrienol

Tocotrienol has several known health benefits including reducing cholesterol, protecting the heart against ischemia, inhibiting cancer tumor growth, lowering blood pressure, anti-aging and uv-protection, and as a potent natural super-antioxidant.
